GM bets on China models for India

Calcutta, Oct. 14: GM India is targeting a double-digit market share with its new models, which are being developed with Chinese joint venture partner Shanghai Automotive Industry Corporation (SAIC). Come November, and the first car from the GM-Shanghai Automotive garage ? the Sail U-VA ? will hit the road. In early, 2013, the MPV Enjoy ? the second car from the duo ? will be out as well.

The company, which had sold 83,969 units from January to September last year, sold 71,500 odd units during the same period this year, a decline of 17 per cent. For the whole of last year, GM India had sold 1,11,510 units.

On whether the company?s sales will touch 1 lakh this year, GM India managing director Lowell Paddock said, ?We hope to see a surge in our sales at the end of the year when our new products will hit the market. I am not happy with the current sales but we hope the economy will turn around during the festive season as a result of popular sentiments as also the set of reforms introduced by the government.?

While the Sail U-VA will be manufactured in the Talegaon plant in Maharashtra, which also makes the Spark and the Beat, the MPV Enjoy will be rolled out of the Hallol plant in Gujarat, which manufactures the Cruze and the Tavera.

?We have a market share of 3.1 per cent. But that is because we were not present in the volume segment of small diesel cars. But now with the Sail U-VA, we hope to catch up. The small SUV is another growing segment in India and Enjoy will be a volume grosser for us. I am looking at a double-digit market share after we have a presence in these two popular segments,? Paddock said.

He, however, ruled out any new small car below the range of Spark to compete with the Maruti?s Alto 800 or the Hyundai?s Eon.

?No, we are not looking at the A segment. We think that segment will shrink as the market matures and the B2 segment will grow,? said Paddock.

According to Paddock, the Sail U-VA is a global product that has been designed in the Pan Asia Technical Automotive Centre (PATAC), Shanghai. The Sail is the number one hatchback in China, Chile and Ecuador.

With the launch of the new U-VA from the GM-SAIC joint venture, China can turn into a hub for GM?s global operations both as a major market and as a design centre for the company's South Asia operations.

(AP) ? Norodom Sihanouk, the revered former king who was a towering figure in Cambodian politics through a half-century of war, genocide and upheaval, died Monday. He was 89.

Sihanouk abdicated the throne in 2004, citing his poor health. He had been getting medical treatment in China since January and had suffered a variety of illnesses, including colon cancer, diabetes and hypertension.

Prince Sisowath Thomico, a royal family member who also was Sihanouk's assistant, said the former king suffered a heart attack at a Beijing hospital.

"His death was a great loss to Cambodia," Thomico said, adding that Sihanouk had dedicated his life "for the sake of his entire nation, country and for the Cambodian people."

Sihanouk's successor, Norodom Sihamoni, is expected to fly to Beijing on Monday to retrieve his father's body, Thomico said.

Cambodian government spokesman Khieu Kanharith said an official funeral will be held once the former king's body is repatriated.

In January, Sihanouk requested that he be cremated in the Cambodian and Buddhist tradition, asking that his ashes be put in an urn, preferably made of gold, and placed in a stupa at the country's Royal Palace.

Sihanouk saw Cambodia transform from colony to kingdom, U.S.-backed regime to Khmer Rouge killing field and foreign-occupied land to guerrilla war zone ? and finally to a fragile experiment with democracy.

He was a feudal-style monarch who called himself a democrat. He was beloved by his people but was seldom able to deliver the stability they craved through decades of violence.

Born on Oct. 31, 1922, Sihanouk enjoyed a pampered childhood in French colonial Indochina.

In 1941, the French crowned 19-year-old Sihanouk rather than relatives closer in line to the throne, thinking the pudgy, giggling prince would be easy to control. They were the first of many to underestimate him, and by 1953 the French were out.

Two years later, Sihanouk stepped down from the throne, organized a mass political party and steered Cambodia toward uneasy neutrality at the height of the Cold War.

Sihanouk accepted limited U.S. aid and nurtured relations with Communist China. He was also a founder of the Non-Aligned Movement.

Sihanouk was a ruthless politician, talented dilettante and tireless playboy, caught up in endless, almost childlike enthusiasms.

He made movies, painted, composed music, fielded a palace soccer team and led his own jazz band. His large appetite extended to fast cars, food and women. He married at least five times ? some say six ? and fathered 14 children.

After 1960, Sihanouk drifted toward the communist camp, seeking assurances from his powerful neighbors, China and Vietnam, that his country's neutrality would be respected.

In 1965, Sihanouk broke off relations with Washington as U.S. involvement in the Vietnam War shifted into high gear. But by 1969, worried about increasing Vietnamese communist use of Cambodian soil, he made new overtures to the United States and turned against China.

Sihanouk's top priority was to keep Cambodia out of the war, but he could not. U.S. aircraft bombed Vietnamese communist sanctuaries in Cambodia with increasing regularity, and his protests were ignored.

Internally, Cambodia was a one-man show. Sihanouk's sharpest critics accused him of running a medieval state as an ancient Khmer ruler reincarnated in Western dress.

"I am Sihanouk," he once said, "and all Cambodians are my children."

Nonetheless, the country was at relative peace and some attempts were made to better the life of the peasants, who adored Sihanouk as a near-deity.

Outsiders saw a country of shimmering temples and emerald green rice fields that seemed a chapter from an Oriental fairy tale. But that face of Cambodia would soon vanish.

In 1970, a U.S.-backed coup sent the prince to Beijing for years of lonely, if lavish, exile. Within weeks, war broke out, beginning a systematic destruction of Cambodia that killed millions and impoverished the survivors.

Sihanouk, seeking to regain the throne, joined the Khmer Rouge-dominated rebels after his overthrow. They had numbered only a few hundred until then, but his presence gave them a legitimacy they had never before enjoyed.

The alliance left Sihanouk open to subsequent criticism that he opened the way for the Khmer Rouge holocaust. But his relations with the rebels were always strained.

"The Khmer Rouge do not like me at all, and I know that. Ooh, la, la ... It is clear to me," he said in a 1973 interview. "When they no longer need me, they will spit me out like a cherry pit."

When the Khmer Rouge seized power in 1975 and Sihanouk returned home, they detained him and ordered his execution. Only the personal intervention of Chinese leader Zhou Enlai saved him.

With Sihanouk under house arrest in the Royal Palace, the Khmer Rouge ran an ultra-radical Maoist regime from 1975 to 1979, emptying the cities to create a vast forced labor camp. An estimated 1.7 million Cambodians were executed or died of disease and hunger under their rule.

Vietnam invaded Cambodia in December 1978 and toppled the Khmer Rouge a few weeks later. Freed as the Vietnamese advanced on Phnom Penh, Sihanouk found exile in Beijing and North Korea.

From there, he headed an unlikely coalition of three guerrilla groups fighting the Vietnamese-installed puppet government. The war lasted a decade.

In a mix of politics and theater ? bringing his French poodle to negotiations, singing love songs over elaborate dinners ? Sihanouk engineered a cease-fire and moves toward national unity and peace.

Sihanouk headed the U. N.-supported interim structure that ran Cambodia until the 1993 elections, lending his prestige to attempts to unite Cambodia's factions.

The election was won by the royalist FUNCINPEC party of Sihanouk's son Prince Norodom Ranariddh. But it was forced into a coalition with the Cambodian People's Party of former Khmer Rouge officer Hun Sen.

In September 1993, Sihanouk re-ascended the throne in a traditional Khmer coronation.

But the bright promise of the elections soon faded.

Four years after the polls, Hun Sen ended his constant bickering with Ranariddh by overthrowing the prince in a violent coup that shattered the results of the election.

International pressure forced Hun Sen to accept Ranariddh's return for a second election in 1998, which was narrowly won by Hun Sen, but ended in more bloodshed as the royalists and other opposition parties forced a constitutional crisis by refusing to join a coalition with the CPP.

Sihanouk stayed on the sidelines for most of the two-year crisis, but as demonstrators clashed in the streets of Phnom Penh, he finally intervened by urging Ranariddh to accept a new coalition with his enemy Hun Sen.

During his last years, Sihanouk's profile and influence receded. While old people in the countryside still held him in reverence, the young generation regarded him as a figure of the past and one partly responsible for Cambodia's tragedy.

Rarely at a loss for words, he became for a time a prolific blogger, posting his musings on current affairs and past controversies. Most of his writing was literally in his own hand ? his site featured images of letters, usually in French in a cramped cursive script, along with handwritten marginalia to news clippings that caught his interest.

His production tailed off, however, as he retreated further from the public eye, spending more and more time under doctor's care in Beijing.

The hard-living Sihanouk had suffered ill health since the early 1990s. He endured cancer, a brain lesion and arterial, heart, lung, liver and eye ailments.

Ailing and weary of politics, Sihanouk stepped down from the throne in 2004 in favor of Sihamoni, a well-liked personality but one with little of the experience needed to negotiate Cambodia's political minefields.

Senior officials in Hun Sen's party were said to favor Sihamoni, a one-time ballet dancer and cultural ambassador, rather than a more combative figure to sit atop the influential throne.

In late 2011, on his return from another extended stay in China, Sihanouk dramatically declared that he never intended to leave his homeland again. But true to his mercurial reputation, he flew off to Beijing just a few months later for medical care.

During the same period, some of the defendants at Cambodia's U.N.-assisted genocide trial of former senior Khmer Rouge figures sought to divert blame from themselves by suggesting that Sihanouk, as their collaborator, shared responsibility for their actions, despite his powerlessness as their virtual prisoner.

HPV shots don't make girls promiscuous, study says

CHICAGO (AP) ? Shots that protect against cervical cancer do not make girls promiscuous, according to the first study to compare medical records for vaccinated and unvaccinated girls.

The researchers didn't ask girls about having sex, but instead looked at "markers" of sexual activity after vaccination against the sexually transmitted human papillomavirus, or HPV. Specifically, they examined up to three years of records on whether girls had sought birth control advice; tests for sexually transmitted diseases or pregnancy; or had become pregnant.

Very few of the girls who got the shots at age 11 or 12 had done any of those over the next three years, or by the time they were 14 or 15. Moreover, the study found no difference in rates of those markers compared with unvaccinated girls.

The study involved nearly 1,400 girls enrolled in a Kaiser Permanente health plan in Atlanta. Results were published online Monday in Pediatrics.

Whether vaccination has any influence on similar markers of sexual activity in older teens wasn't examined in this study but other research has suggested it doesn't.

The study is the first to use medical outcomes data to examine consequences of HPV vaccination and the results are "comforting and reassuring," said lead author Robert Bednarczyk, a researcher at Kaiser and Emory University. Both institutions paid for the study.

HPV is the leading cause of cervical cancer and also has been linked with anal and oral cancers in women and men.

The federal Centers for Disease Control and Prevention and the American Academy of Pediatrics recommend HPV shots for girls and boys at age 11 or 12, before they have ever had sex. Three doses are generally recommended over six months.

Some parents have raised concerns that the shots "are a license to have sex," but the study bolsters evidence against that concern, said Dr. Elizabeth Alderman, an adolescent medicine specialist at The Children's Hospital at Montefiore in New York City. She was not involved in the study.

A CDC study published in January suggested that the shots don't promote sexual activity among older girls, but it relied on self-reporting, at ages 15 to 24. That's a less reliable method than the new study, Alderman said. She has been a paid speaker for Merck & Co., which makes one of the two HPV vaccines sold in the United States, but said she has no current financial ties to the company.

In the new study, at least 90 percent of vaccinated and unvaccinated girls did not seek pregnancy tests, chlamydia tests or birth control counseling, markers that were considered surrogates for sexual activity during up to three years of follow-up. Two in each group became pregnant. Chlamydia, a common sexually transmitted disease, was diagnosed in one vaccinated girl and three unvaccinated girls.

Three of the study's four co-authors reported having done previous research funded by Merck.

Cardinals, Giants each pulled off remarkable rally

SAN FRANCISCO (AP) ? As many times as he gets asked, Cardinals closer Jason Motte still has no perfect answer for how St. Louis found a way to win at Washington after trailing 6-0 and get back to the NL championship series.

"These guys just prove what big hearts they have and how much they go out there and work their butts off," said Motte, Friday night's winning pitcher. "Someone asked me last night how we keep doing it, and I said, 'I don't know, maybe we're just stubborn. We just don't give up.' That's kind of how you have to be."

St. Louis manager Mike Matheny will watch the game again, once things slow down, so he can truly appreciate just what his Cardinals accomplished in beating the Nationals ? the team with baseball's best record this season.

San Francisco skipper Bruce Bochy doesn't need another look to know how impressive the reigning World Series champions' ninth-inning comeback was for a 9-7 victory in the nation's capital.

Bochy's team had its own remarkable rally that's not quite as fresh as the Cardinals' feat: Three road wins at Cincinnati to advance after dropping the first two games of the division series at home to the Reds.

The last two World Series winners sure are showing their championship mettle in mid-October.

They will face off in Game 1 of the NLCS on Sunday night at San Francisco's AT&T Park. Left-hander Madison Bumgarner gets the ball for the Giants at home against 6-foot-5 right-hander Lance Lynn.

Bumgarner, a 16-game winner this year, lost Game 2 of the NL division series here to Cincinnati.

"I felt good last time, things just didn't go my way," Bumgarner said. "That's the way this game is."

While the Giants became the eighth team to come back from a 2-0 deficit in a best-of-five series and first to do it on the road, the Cardinals earned the biggest comeback ever in a winner-take-all postseason game, according to STATS LLC.

"It really hasn't sunk in," Matheny said after an all-night, cross-country trip to the Bay Area. "I see a knockdown-drag out ahead of us. I'm certain Major League Baseball has to be very pleased with the caliber of baseball that's happened so far in this postseason. And I don't see any reason why the excitement wouldn't continue. We're looking at two well-rounded teams."

Daniel Descalso hit a tying, two-out single, and Pete Kozma added a go-ahead, two-run single to lead the Cardinals' rally.

The Giants, all the while, waited out the game on their team plane still parked on the tarmac in Cincinnati. Players gathered around iPads to watch the improbable comeback by a Cardinals club managed by former Giants catcher, Matheny.

The Cardinals have won all six of their games when facing elimination the past two years, down to their final strike not once but twice against the Texas Rangers in last year's World Series before prevailing.

This time, they did it against the team with 98 wins.

"I don't why we do it. We love the dramatics. I'd like us to not keep waiting until the last strike," pitcher Kyle Lohse said, "because that's too much for my heart to handle."

The Giants barely beat St. Louis to San Francisco after getting delayed more than three hours as their plane refueled and dealt with mechanical problems.

Initially, the plane had enough fuel to take the NL West winners to Washington. Instead, the Giants got to come home. They landed at 5:09 a.m.

"Everyone was rooting and cheering for the Cardinals because we wanted to go home," third baseman Pablo Sandoval said. "I was cheering for St. Louis. I was excited to be back home. Probably almost everybody was cheering for the Cardinals. Every guy was happy to be home. Once we were in the air, I slept the whole flight."

Yankees manager Joe Girardi fell asleep before the NL clincher ended. The Cardinals were trailing 7-5 the last thing he remembered. His television was still on, and when he opened his eyes at one point he saw the final score.

"I said, 'What happened?'" he said. "I looked at it, the game was over and I closed my eyes and went back to bed. Pretty amazing. You think about the games, and you are down 6-0 and you're able to come back. And you keep going at it. This first round was probably as good as it gets. Probably as good of baseball as we have seen when you talk about eight teams playing."

Road weary and riding high, the Cardinals and Giants are counting on more close games and playing all 27 outs ? or more.

"It's definitely a fun matchup," said Giants catcher Buster Posey, whose grand slam helped seal Game 5 against the Reds. "These should be some fun games to watch. Both teams have good arms, good offenses. And the resiliency of both teams is the strength of both teams."

Complex logic circuit made from bacterial genes

ScienceDaily (Oct. 12, 2012) ? By force of habit we tend to assume computers are made of silicon, but there is actually no necessary connection between the machine and the material. All that an engineer needs to do to make a computer is to find a way to build logic gates -- the elementary building blocks of digital computers -- in whatever material is handy.

So logic gates could theoretically be made of pipes of water, channels for billiard balls or even mazes for soldier crabs.

By comparison Tae Seok Moon's ambition, which is to build logic gates out of genes, seems eminently practical. As a postdoctoral fellow in the lab of Christopher Voigt, PhD, a synthetic biologist at the Massachusetts Institute of Technology, he recently made the largest gene (or genetic) circuit yet reported.

Moon, PhD, now an assistant professor of energy, environmental and chemical engineering in the School of Engineering & Applied Science at Washington University in St. Louis is the lead author of an article describing the project in the Oct. 7 issue of Nature. Voigt is the senior author.

The tiny circuits constructed from these gene gates and others like them may one day be components of engineered cells that will monitor and respond to their environments.

The number of tasks they could undertake is limited only by evolution and human ingenuity. Janitor bacteria might clean up pollutants, chemical-engineer bacteria pump out biofuels and miniature infection-control bacteria might bustle about killing pathogens.

How to make an AND gate out of genes

The basis of modern computers is the logic gate, a device that makes simple comparisons between the bits, the 1s and 0s, in which computers encode information. Each logic gate has multiple inputs and one output. The output of the gate depends on the inputs and the operation the gate performs.

An AND gate, for example, turns on only if all of its inputs are on. An OR gate turns on if any of its inputs are on.

Suggestively, genes are turned on or off when a transcription factor binds to a region of DNA adjacent to the gene called a promotor.

To make an AND gate out of genes, however, Moon had to find a gene whose activation is controlled by at least two molecules, not one. So only if both molecule 1 AND molecule 2 are present will the gene be turned on and translated into protein.

Such a genetic circuit had been identified in Salmonella typhimurium, the bacterium that causes food poisoning. In this circuit, the transcription factor can bind to the promotor of a gene only if a molecule called a chaperone is present. This meant the genetic circuit could form the basis of a two-input AND gate.

The circuit Moon eventually built consisted of four sensors for four different molecules that fed into three two-input AND gates. If all four molecules were present, all three AND gates turned on and the last one produced a reporter protein that fluoresced red, so that the operation of the circuit could be easily monitored.

In the future, Moon says, a synthetic bacterium with this circuit might sense four different cancer indicators and, in the presence of all four, release a tumor-killing factor.

Crosstalk and timing faults

There are huge differences, of course, between the floppy molecules that embody biological logic gates and the diodes and transistors that embody electronic ones.

Engineers designing biological circuits worry a great deal about crosstalk, or interference. If a circuit is to work properly, the molecules that make up one gate cannot bind to molecules that are part of another gate.

This is much more of a problem in a biological circuit than in an electronic circuit because the interior of a cell is a kind of soup where molecules mingle freely.

To ensure that there wouldn't be crosstalk among his AND gates, Moon mined parts for his gates from three different strains of bacteria: Shigella flexneri and Pseudomonas aeruginosa, as well as Salmonella.

Although the parts from the three different strains were already quite dissimilar, he made them even more so by subjecting them to error-prone copying cycles and screening the copies for ones that were even less prone to crosstalk (but still functional).

Another problem Moon faced is that biological circuits, unlike electronic ones, don't have internal clocks that keep the bits moving through the logic gates in lockstep. If signals progress through layers of gates at different speeds, the output of the entire circuit may be wrong, a problem called a timing fault.

Experiments designed to detect such faults in the synthetic circuit showed that they didn't occur, probably because the chaperones for one layer of logic gates degrades before the transcription factors for the next layer are generated, and this forces a kind of rhythm on the circuit.

Hijacking a bacterium's controller

"We're not trying to build a computer out of biological logic gates," Moon says. "You can't build a computer this way. Instead we're trying to make controllers that will allow us to access all the things biological organisms do in simple, programmable ways."

"I see the cell as a system that consists of a sensor, a controller (the logic circuit), and an actuator," he says. "This paper covers work on the controller, but eventually the controller's output will drive an actuator, something that will do work on the cell's surroundings. "

An synthetic bacterium designed by a friend of Moon's at Nanyang Technological University in Singapore senses signaling molecules released by the pathogen Pseudomonas aeruginosa. When the molecules reach a high enough concentration, the bacterium generates a toxin and a protein that causes it to burst, releasing the toxin, and killing nearby P. aeruginosa.

"Silicon cannot do that," Moon says.
