Older Fathers Pass On More Mutations to Children

Risks for autism and schizophrenia rise for kids of older fathers, and a new genetic study suggests why

In the 1930s, the pioneering geneticist J. B. S. Haldane noticed a peculiar inheritance pattern in families with long histories of haemophilia. The faulty mutation responsible for the blood-clotting disorder tended to arise on the X?chromosomes that fathers passed to their daughters, rather than on those that mothers passed down. Haldane subsequently proposed1 that children inherit more mutations from their fathers than their mothers, although he acknowledged that ?it is difficult to see how this could be proved or disproved for many years to come?.

That year has finally arrived: whole-genome sequencing of dozens of Icelandic families has at last provided the evidence that eluded Haldane. More?over, a study published in Nature findsthat the age at which a father sires children determines how many mutations those offspring inherit2. By starting families in their thirties, forties and beyond, men could be increasing the chances that their children will develop autism, schizophrenia and other diseases often linked to new mutations. ?The older we are as fathers, the more likely we will pass on our mutations,? says lead author K?ri Stef?nsson, chief executive of deCODE Genetics in Reykjavik. ?The more mutations we pass on, the more likely that one of them is going to be deleterious.?

Haldane, working years before the structure of DNA was determined, was also correct about why fathers pass on more mutations. Sperm is continually being generated by dividing precursor cells, which acquire new mutations with each division. By contrast, women are born with their lifelong complement of egg cells.

Stef?nsson, whose company maintains genetic information on most Icelanders, compared the whole-genome sequences of 78?trios of a mother, father and child. The team searched for mutations in the child that were not present in either parent and that must therefore have arisen spontaneously in the egg, sperm or embryo. The paper reports the largest such study of nuclear families so far.

Fathers passed on nearly four times as many new mutations as mothers: on average, 55 versus 14. The father?s age also accounted for nearly all of the variation in the number of new mutations in a child?s genome, with the number of new mutations being passed on rising exponentially with paternal age. A 36-year-old will pass on twice as many mutations to his child as a man of 20, and a 70-year-old eight times as many, Stef?nsson?s team estimates.

The researchers estimate that an Icelandic child born in 2011 will harbour 70 new mutations, compared with 60 for a child born in 1980; the average age of fatherhood rose from 28 to 33 over that time.

Most such mutations are harmless, but Stef?nsson?s team identified some that studies have linked to conditions such as autism and schizophrenia. The study does not prove that older fathers are more likely than younger ones to pass on disease-associated or other deleterious genes, but that is the strong implication, Stef?nsson and other geneticists say.

Previous studies have shown that a child?s risk of being diagnosed with autism increases with the father?s age. And a trio of papers3?5 published this year identified dozens of new mutations implicated in autism and found that the mutations were four times more likely to originate on the father?s side than the mother?s.

"Real Housewives of New Jersey" cast member Jacqueline Laurita has revealed that her youngest child, son Nicholas, now 3, has been diagnosed with autism.

In an interview with People magazine, Laurita?says that?Nicholas started exhibiting worrisome behavior at around 18 months. He began regressing with his speech and motor skills, then refusing to answer to?his name or notice other people in the room.

"We had no idea what was going on," Laurita said. Amid the uncertainty, the "Housewives" star said she hid her son's medical issues from cameras as she taped the fourth season of the show, which is airing now.

Laurita, who is also mom to Ashlee, 21, and C.J., 10, said that she's concerned about what the future holds for Nicholas.

"I worry about him being independent when he's older. I spend all my time researching what we can do for him," she told the magazine.

"You never want to think that your child isn't perfectly healthy," Laurita's husband Chris told People. "We didn't want to believe it was true."

The Arginine Paradox ? find out if arginine based NO boosters really work for you

Some of the most popular supplements today are the so called pre-workout nitric oxide (NO) boosters 1, 2. These contain a panoply of ingredients, but the main one is arginine. The rationale goes that arginine is a nitric oxide (NO) precursor and NO is a potent vasodilator 3, 4, which in turn supposedly will boost blood flow to exercising muscles, performance and recovery. And while arginine supplementation is beneficial for various clinical populations (see below), studies in healthy adults have not unequivocally supported the marketing hype surrounding arginine supplementation and nitric oxide boosters 1, 5, 6. Why? Let?s take a look under the hood?

Some reasons explaining the inefficacy of arginine supplementation and nitric oxide boosters

The popular pre-workout NO boosting supplements rely on the arginine-NO pathway and a lot of theoretical assumptions. Some reason for the conflicting findings on arginine supplementation are that studies have used different routes of administration (oral vs. intravenous), various forms of L-arginine, varying exercise testing protocols, different test subjects (trained vs. untrained, young vs. elderly), and supplement cocktails containing other substances (eg. creatine, beta-alanine, caffeine, carbs etc) that indeed do have performance enhancing effects.

The rationale for L-arginine supplementation is based largely on research using intravenous L-arginine, often at dosage of 30 g. This has no practical relevance since most, if not all, supplements are taken orally. In a direct head-to-head comparison of oral and intravenous L-arginine administration, no effect on vasodilatation was found after oral L-arginine supplementation 7. One reason for this could be the extensive elimination of orally ingested L-arginine due to intestinal arginase activity and low bioavailability 8. This would require intake of very large amounts of L-arginine, which not only has an unpleasant taste, but also can cause gastric problems 9.

The Arginine Paradox

The backbone running the arginine-NO pathway is an enzyme called eNOS (endothelial nitric oxide synthase) 10. One factor that affects the velocity of an enzyme catalyzed reaction is the concentration of the substance the enzyme uses (a.k.a substrate). L-arginine is a substrate for the NOS, which converts L-arginine into NO 11. For eNOS, the Michaelis-Menten (Km) constant (which is nerd speak denoting the substrate concentration with which half the enzyme reaction velocity maximum rate is attained) is about 3 mmol/L 12, whereas the blood L-arginine concentrations in both healthy and diseased individuals ranges from 40 to 100 mmol/L 11. This means that the typical non-supplemented blood level of is high enough to saturate endothelial NOS. And when an enzyme is saturated with substrate (in this case L-arginine) more substrate won?t have an effect on a reaction. Therefore, if you?re healthy, when you supplement L-arginine (even in large dosages) you won?t get more NO from the arginine-NO pathway, because L-arginine is not rate-limiting for eNOS.

However, the story is different in clinical conditions like e.g. high blood pressure 13, 14, elevated cholesterol levels 15, 16, heart disease 17, insulin resistance 18, diabetes 19, 20 and in the elderly population 14, 21, in whom L-arginine supplementation has beneficial effects, possibly due to increased NO production 14. Thus, it appears that L-arginine is a limiting factor for NO synthesis in clinical conditions, but not for healthy individuals.

The term ?L-arginine paradox? refers to specific situations in which L-arginine supplementation indeed does stimulate NOS activity and NO production, even when blood arginine levels are within the normal range. One explanation for the L-arginine paradox is the presence of high levels of ADMA (asymmetric dimethylarginine), which is an inhibitor of eNOS 10, 22-24. ADMA is produced as part of the body?s normal metabolism 25-27, but in clinical conditions the ADMA levels are elevated several fold 24. In the presence of elevated blood levels of ADMA, eNOS activity diminishes, resulting in a reduced NO production.

How L-arginine supplementation works in the context of elevated ADMA levels

Because ADMA ?displaces? L-arginine and thereby reduces the L-arginine availability for eNOS, the ratio of L-arginine to ADMA determines how much of the L-arginine that is floating around in the blood that will be available for eNOS to use for NO production 28, 29. In conditions with elevated ADMA levels, L-arginine supplementation will antagonize ADMA by re-establishing the L-arginine/ADMA ratio and thereby ?re-activating? eNOS and increasing NO production 30. This is supported by studies showing that L-arginine supplementation reverses endothelial dysfunction attributable to high ADMA levels in clinical populations 21, 31-33, and that there exist a direct correlation between the change in L-arginine/ADMA ratio and the change in blood flow mediated dilation (vasodilatation) 8.

How to combat elevated ADMA levels

The importance of ADMA is underscored by the fact that it is considered a novel cardiovascular risk factor 22, 33, 34. ADMA seems to mediate the effect of many risk factors on the NOS pathway. Therefore, blood ADMA levels have been suggested to be an ??ber marker?, or an overall risk factor that reflects the summative effect of all risk factors on endothelial and cardiovascular health 26.

Because of this, drug companies are currently fervently trying to develop drugs that lower ADMA levels. As of this writing there is no ADMA specific drug available. The strongest candidate as an ADMA specific drug is DDAH, the enzyme that naturally breaks down ADMA 27, 35, 36, and whose activity is also impaired in the above mentioned clinical conditions. DDAH boosting drugs are still in the research phase of development, and it will likely take many years before they, or any other ADMA lowering drugs hit the market. However, several well known drugs used for diabetes (metformin, rosiglitazone) and high blood pressure (ACE inhibitors, angiotensin receptor antagonists) have been shown to lower ADMA levels 37-40. Today, the only dietary option is L-arginine supplementation for those in need.

Will L-arginine supplementation benefit me?

If you are insulin resistant (have elevated levels of insulin and/or blood glucose), have high blood pressure, elevated blood cholesterol, high homocysteine levels 32, diabetes or cardiovascular disease, and/or passed your middle-age a while ago, L-arginine supplementation will work for you in restoring subpar NO production.

In contrast, if you don?t have any risk factors and, save your money. In the next article I will cover a less well known NO producing pathway, that can benefit both healthy and risk factor affected folks.

Harness, MD: IORT radiation therapy has been around for a longtime, but the resurgence in the interest in intraoperative radiation therapy is the fact that we now have mobile linear accelerators that can deliver very high doses of radiation therapy directly to where the cancer was at the time of surgery in an operating room that is not a big fixed cement vault that we generally need for a linear accelerator.? IORT has been of big use and big boom in Europe and is now coming to the United States. ?Here in my practice at St. Joseph Hospital we are really lucky to have a mobile linear accelerator called the Mobetron. We have been using this technology for the past three years to do the boost part of whole breast radiation therapy starting the radiation therapy at the time of surgery so we can directly look right onto the area where the cancer was, making sure the dose of the radiation therapy is being properly delivered.? We also have two protocols in addition here at St. Joseph Hospital where we do the boost intraoperatively followed by a shortened hypofraction radiation therapy or we now have a single fraction one time only radiation therapy in selected cases and again that is done through a research protocol.? So clearly IORT is cutting edge treatment of breast cancer.? I am very fortunate to have it available for my practice.

NEW YORK (AP) ? President Barack Obama sees the presidential contest as a clutch moment in his favorite sport ? the fourth quarter of a taut basketball game.

Fundraising with the help of current and former National Basketball Association stars, Obama told a small group of donors gathered Wednesday night at Alice Tully Hall in Lincoln Center that the current campaign is like the final minutes of a basketball contest with his team up by a few points.

"But the other side is coming strong," Obama said. "And they play a little dirty. We've got a few folks on our team in foul trouble. We have a couple of injuries. And I believe they have one last run in them."

Obama raised $3 million with the help of the players, who included former Chicago Bulls player Michael Jordan and former New York Knicks Walt Frazier and Bill Bradley. NBA Commissioner David Stern also was there.

Invoking Jordan's competitive nature, Obama concluded, "If you have seven minutes to go and you have a little bit of a lead, that's when you put them away."

The players were part a daylong fundraiser. Earlier, former and current players participated in a $250-per-person autograph session and in a skills camp priced at $5,000 for two people.

Well, where I am it's 5am, and I technically leave the house in about 6 hours...so I really should be in bed to be honest XD
And I'll be back in 3 or 4 days, if I've calculated the time right :) But I also might end up getting internet connection inside my hotel room, which will mean it won't be an issue and I'll be able to edit my CSs and send them in whenever :)

It's mostly the male characters that I'm concerned about as those tend to be the ones people forget. Well, I'm gonna get some sleep now, and hopefully if I have time in the morning between packing and anything, I'll get them done. If I can't get the whole thing bulked up, can I just make a start on both CSs, writing in little details and submitting them, and then giving more detail and everything when I get back or that night if I get internet connection?

A crisp is a pie without the fuss of a crust.

This humble cousin of upper-crust pies and tarts melds tree-ripened fruit and a few pantry staples into a sweet-tart old-fashioned dessert that?s hard to resist.

The British call it a crumble. Americans call it a crisp. We call it downright delicious.

All those hot fruit juices bubble up into the buttery, sugary topping as it bakes to create sophisticated flavors that are mouth-watering and good.

Crisps are so easy to assemble. Mix fruit, sugar, lemon juice and tapioca and pour into a deep baking dish.

Sprinkle on a crumbly topping to create a one-of-a-kind dessert. Change up the fruit, using whatever is on hand. Mix apricot and pineapple or pineapple juice to lighten up an otherwise heavy filling. A mix of fresh cherries and canned cherry pie filling produces excellent results. Firm pears are an unexpected surprise. Add a few tablespoons of apricot jam to deepen their flavor.

Use whatever thickener you have on hand. Flour will do in a pinch. Even better is arrowroot, cornstarch or potato starch. Quick-cook tapioca, though, is my standby. It produces a clear filling that lets the fruit flavors shout and the brightly colored fillings shine. Swap brown sugar for white sugar or use a mix of both in the topping or the filling. Sprinkle in a little cinnamon, nutmeg or cloves into the topping. A 1/4 teaspoon of each are all you?ll need. Add a dash of spice to the filling. Or don?t. This rustic dessert takes kindly to improvisation and is kind to the hostess because it comes together effortlessly. And if at first bite it?s a little too tart, make room for ice cream. A tart crisp and vanilla ice cream are heaven in a bite.

Change up the topping: Try oatmeal or ground nuts; just butter, sugar and flour work great; and finely chopped nuts in a butter-flour-sugar topping are wonderful because they toast during baking and provide flavor and texture.

Pop it in the oven for a bit. When those delicious fruit juices bubble up over the filling, you know it?s done. There?s no guesswork here, like with a pie filling under a crust.

Resist the urge to eat it hot out of the oven ? the juices need to thicken and set. And if you?re lucky enough to have leftovers, you?ll be rewarded with flavors that are even better the second day.

Blueberry-Peach Crisp

5 cups frozen blueberries, unthawed

2/3 cup granulated sugar

1/4 cup plus 2/3 cup water

1/4 cup cornstarch

Grated zest of 1 lemon

2 medium ripe peaches, peeled, pitted and sliced

Preheat oven to 400 degrees.

In a large saucepan, combine the blueberries, granulated sugar and 1/4 cup water. Bring to a simmer over medium heat, stirring occasionally. Whisk together the cornstarch and remaining 2/3 cup water in a small bowl until smooth. Stir in cornstarch mixture into the hot berries. Gently stir in lemon zest and peaches, be careful not to mash the peaches.

Reduce the heat to low and continue simmering the fruit, gently stirring, until the juices have thickened and the mixture is clear. Remove from heat and scoop mixture into 2-1/2-quart baking dish.

Bake the crisp until the topping is nicely browned, 30 to 40 minutes.

The suggested topping for this crisp combines 3/4 cup all-purpose flour, 1/2 cup old-fashioned rolled oats, 1/2 cup firmly packed brown sugar, 1 teaspoon nutmeg and 1/4 cup nonhydrogenated margarine (stick with butter, though, its flavor can?t be beat). The ingredients are combined and sprinkled on the fruit.

Blackberry Crisp

4 cups frozen blackberries

1-1/2 to 1-2/3 cups granulated sugar

3 tablespoons cornstarch

1/2 teaspoon cider vinegar

2 tablespoons water

1 tablespoon quick-cooking tapioca

In a heavy-bottom saucepan, combine berries, sugar, cornstarch, vinegar and 2 tablespoons water. Mash the fruit slightly. Cook over medium-low heat, stirring occasionally, just until mixture nears the boiling point. Remove from heat. Stir in tapioca. Bake at 350 degrees for 30 to 40 minutes. The cider vinegar softens the seeds.

Alternately, mix blackberries when they?re in season with the sugar and 3 to 4 tablespoons tapioca. Omit the cornstarch, cider vinegar and water. Freeze until ready to bake. Thaw slightly, add crumble topping and bake until bubbly. Freezing also soften the seeds.

Nut Topping

2/3 cup granulated sugar

7 tablespoons butter, room temperature

1/2 cup plus 2 tablespoons all-purpose flour

1 cup chopped walnuts, pecans or almonds

1/2 teaspoon each cinnamon and nutmeg

Combine all ingredients and crumble with your fingertips. Spread mixture over prepared fruit in buttered baking dish and bake as directed.

Oatmeal Topping

3/4 cup all-purpose flour

1 cup packed light brown sugar

1-1/2 cups rolled oats

6 tablespoons ( 3/4 stick) salted butter, softened

Combine dry ingredients in a bowl and mix. Add butter and mix by hand until butter is incorporated. Spread over fruit. Bake until top is golden, 40 to 45 minutes.

For the filling, Lauren combines 3 cups blueberries, 2 tablespoons sugar and the juice of 1 lemon as the base for this crisp. She bakes it in a 9-inch square baking dish. She also doesn?t use a thickener.

Change up the topping by reducing sugar and oatmeal by half and flour to 1/3 cup. Add 1/4 cup toasted wheat germ.

Ground Nut Topping

3/4 cup sugar

1-1/4 cups all-purpose flour

3/4 cup almond meal

3/4 cup butter, softened

6 tablespoons sliced almonds, divided use

Mix the dry ingredients in a bowl, except 3 tablespoons of the sliced almonds.

Add the butter and rub with your hands until the mixture turns crumbly. Sprinkle topping on fruit and scatter the remaining sliced almonds on top.

Preheat oven to 350 degrees. Bake 40 minutes or until fruit is bubbly and the topping browned.

Henry uses no thickener in her fruit filling: 1-1/3 pounds apricots, quartered and pitted; 3 peaches, pitted and sliced; 1 pound blackberries; zest of half lemon and juice of entire lemon; and 3tablespoons sugar.

Peach Filling

5 to 6 cups peaches

Juice of 1 lemon

1/2 to 3/4 cup granulated sugar

3 tablespoons quick-cooking tapioca, increase if fruit is especially juicy

Note: Use this recipe as a starting point for the fruit filling in crisps, just adjust the sugar to suit the sweetness of the fruit.

Combine filling ingredients. Sprinkle with a crumb topping. Bake at 350 degrees for 35 to 45 minutes.
